SNAP-8 (Acetyl Octapeptide-3)
Also known as: Leuphasyl, Acetyl Octapeptide-3, SNAP-8 peptide, Argireline extension, Acetyl Glutamyl Heptapeptide-3
SNAP-8 is a cosmetic peptide active. Not a drug; for topical use only.
Overview
8-amino acid cosmetic peptide active (Acetyl Octapeptide-3) that competes with SNARE complex proteins to reduce neuromuscular signal transmission in facial expression muscles. An extended, more potent version of Argireline (Acetyl Hexapeptide-3). Clinical studies show 26–63% reduction in expression wrinkle depth with twice-daily application, positioning it as a topical botulinum toxin alternative.
Research Summary
SNAP-8 mimics the N-terminal domain of SNAP-25, competing for SNARE complex assembly required for acetylcholine vesicle fusion at the neuromuscular junction. This partial inhibition reduces muscle contraction intensity without complete paralysis. Clinical studies demonstrate statistically significant reductions in crow's feet and forehead line depth after 28 days of use at 5–10% concentration.

SNAP-8 (Acetyl Octapeptide-3, also known as Acetyl Glutamyl Heptapeptide-3) is a synthetic octapeptide designed as an extended analogue of Argireline (Acetyl Hexapeptide-3). Both compounds share the same fundamental mechanism — interference with SNARE complex assembly at the neuromuscular junction — but SNAP-8's additional two amino acids provide enhanced receptor engagement and stability.
The compound was developed with the goal of creating a topical alternative to botulinum toxin for expression wrinkles — fine lines formed by repeated facial muscle contraction over time. While it cannot achieve the complete paralysis of Botox, its partial inhibition approach is favored for topical cosmetic use where some expression retention is desirable.
Mechanism
SNARE Complex Interference
The neuromuscular junction signal cascade that SNAP-8 targets:
- Motor nerve depolarization → voltage-gated calcium channels open → Ca²⁺ influx
- SNARE complex assembly: VAMP-2 (vesicle membrane) + Syntaxin (cell membrane) + SNAP-25 (cell membrane) form a 4-helix bundle that pulls membranes together
- Vesicle fusion: Acetylcholine-containing vesicle fuses with presynaptic membrane
- ACh release → binds nicotinic receptors on muscle → contraction
SNAP-8 intervention point: The peptide mimics the N-terminal domain of SNAP-25. By competing for the SNARE complex binding site:
- Partial occupancy of SNARE sites reduces vesicle fusion frequency
- Less acetylcholine released per nerve impulse
- Reduced muscle contraction amplitude
- Gradual expression wrinkle reduction with continued use
Why It's Not Botox
Botulinum toxin cleaves SNAP-25 enzymatically — complete, irreversible inactivation. SNAP-8 is a competitive peptide that merely occupies binding sites: reversible, dose-dependent, partial effect. This is why SNAP-8 shows 26–63% wrinkle reduction (not 90%+ like Botox) but also has an excellent safety profile with no systemic effects.
SNAP-8 vs. Argireline
| Parameter | Argireline (Acetyl Hexapeptide-3) | SNAP-8 (Acetyl Octapeptide-3) | |-----------|----------------------------------|-------------------------------| | Length | 6 amino acids | 8 amino acids | | SNAP-25 mimicry | Partial | More complete | | Efficacy | ~30% wrinkle reduction | ~26–63% wrinkle reduction | | Typical concentration | 3–5% | 3–10% | | Cost | Lower | Higher | | Stability | Good | Good |
Formulation Notes
SNAP-8 is best incorporated into light serums or hydrogels that maximize skin contact time at the target concentration. Heavier creams may dilute effective concentration. Application to clean, slightly damp skin improves peptide absorption. Typical protocol: apply twice daily to expression areas (crow's feet, forehead, glabellar lines) for minimum 28 days before evaluating results.
