MGF vs Mod-GRF(1-29)
A neutral, side-by-side comparison of two performance & growth hormone peptides — their mechanisms, dosing ranges, administration routes, side effects, and research evidence. For research and educational purposes only.
MGF
MGF (Mechano Growth Factor) is a splice variant of IGF-1 produced locally in muscle tissue in response to mechanical loading (exercise, especially eccentric loading). Unlike circulating IGF-1 which is produced primarily by the liver, MGF is generated within the muscle itself and acts locally to activate satellite cells (muscle stem cells) — the cells responsible for muscle repair, growth, and adaptation. It is considered the primary local anabolic signal generated by resistance training.
Full profileMod-GRF(1-29)
Mod-GRF(1-29) is a stabilized analog of growth hormone-releasing hormone (GHRH) that spans residues 1–29 of the native peptide. Unlike CJC-1295 (which includes a Drug Affinity Complex for multi-day half-life), Mod-GRF(1-29) has a short ~30-minute active window — mimicking the natural GHRH pulse pattern. It is the preferred GHRH component for precise, pulsatile GH protocols when combined with a GHRP.

Key differences
- Evidence level differs: MGF is preclinical only, while Mod-GRF(1-29) is research-stage.
- Administration: MGF — Intramuscular injection (into worked muscle — critical for local effect), Subcutaneous injection (produces more systemic distribution); Mod-GRF(1-29) — Subcutaneous injection, Intramuscular injection.
- Frequency: MGF is typically Post-workout (within 30–60 minutes of training), 3–4x weekly; Mod-GRF(1-29) is 2–3x daily, co-administered with a GHRP (pre-sleep, pre-workout, and/or upon waking).
- Research depth: this profile cites 3 sources for MGF vs 2 for Mod-GRF(1-29).
How each works
MGF
MGF is encoded by the IGF-1 gene but produced via alternative splicing that generates a unique 49-amino acid C-terminal E-peptide (Ec peptide). The Ec peptide has distinct biological activity from the IGF-1 domain — it directly activates satellite cell proliferation through a receptor pathway separate from the standard IGF-1R. Studies in rodents show that local MGF injection produces significant muscle hypertrophy in the injected limb without systemic IGF-1 elevation. MGF expression spikes within 30 minutes of resistance exercise and declines over 24–48 hours — aligning precisely with the post-exercise anabolic window.
Mod-GRF(1-29)
Native GHRH(1-29) is rapidly cleaved by plasma dipeptidyl peptidase IV (DPP-IV) within 2–3 minutes. Mod-GRF(1-29) incorporates four amino acid substitutions (positions 2, 8, 15, 27) that resist DPP-IV degradation, extending the half-life to approximately 30 minutes while preserving GHRH receptor binding affinity. This creates a half-life window aligned with physiological GH pulsatility, making it suitable for 2–3x daily injections that mimic natural GH release patterns.
